Do I need both, a reseller certificate which I have, and a business license when I dropship or work at home online with Ebay or my online store?

A business license is a local issue. In most cases it is mandatory.

Additionally if you are trading under any name other than your own name you will need to file a fictitious name certification (sometimes called a "DBA") with the local city or county offices.
